DESCRIPTION:“Single-cell to tissue-scale memory of infection and inflammation” \nThe role of epithelial and stromal cells in driving inflammation\, together with immune cells\, is being increasingly appreciated. However\, we currently lack tissue-scale frameworks to define and test how inflammation collectively impacts epithelial\, stromal\, and immune cells. This poses a fundamental challenge for understanding the cellular mechanisms that drive or sustain health and disease within\, and across\, barriers such as the skin\, airway\, and intestine. Our lab is addressing this challenge by quantifying and controlling how inflammation changes the ecology of barrier tissues. Our central hypothesis is that inflammation is distributed and stored in multiple cell types\, with the capacity to form tissue-level collective cellular networks. Starting from our human single-cell RNA-sequencing data collected from infectious and inflammatory diseases\, we are developing quantitative approaches to define how static cellular ecosystems are arranged in barrier tissues. These studies are informing time-series modeling of murine inflammatory models focused on Type 1\, 2 and 17 inflammation to understand how the same pressure applied to epithelial\, stromal\, and immune cell subsets shifts functional states and interactions in distinct barrier tissues. We anticipate to identify the gradual inflammatory transitions which maintain essential tissue functionality and critical “tipping points” in tissue state that compromise barrier functions.

DESCRIPTION:Maternal diet quality and health status of newborns\nLecturer: Mgr. Jitka Pavlíková\, Ph.D. / Department of Toxicology a Molecular Epidemiology \nAnnotation: Persistent organic pollutants are compounds of mostly anthropogenic origin that persist in the environment for a long time\, become part of food chains\, and some of them accumulate in living organisms. These substances are being monitored for their adverse effects on organisms\, and the use of some of them has already been banned or at least significantly restricted. In our study\, we analyzed possible associations between the health status of newborns (birth weight\, oxidative damage levels) and maternal diet. We obtained information about diet quantity and quality from the maternal diet records and determined the concentrations of 67 different persistent organic pollutants in the collected samples. \nThe seminar will take place on 4. 4. 2024 from 2 p.m. in the Turquoise Auditorium of the IEM CAS. \nWe look forward to your participation.

DESCRIPTION:“Chromatin ADP-ribosylation in neurodegenerative disease” \nADP-ribosylation is a common protein post-translational modification that regulates numerous cellular processes\, including transcription\, cell proliferation and differentiation\, immune and stress responses\, aging\, and cell death. One of its most extensively studied roles is in DNA damage response and subsequent DNA repair. Moreover\, deregulation of ADP-ribose metabolism is linked to neurological and neurodevelopmental disorders. Interestingly\, our recent work reveals that serine ADP-ribosylation is largely dispensable for DNA repair. We propose that deregulated serine ADP-ribosylation might instead lead to alterations in other histone modifications\, changes in chromatin remodeling and gene expression\, and consequently could contribute to the development of neurological diseases.

DESCRIPTION:“Molecular mechanisms of proteoglycan-mediated semaphorin signaling in axon guidance” \nThe precise assembly of a functional nervous system relies on axon guidance cues. Beyond engaging their cognate receptors and initiating signaling cascades that modulate cytoskeletal dynamics\, guidance cues also bind components of the extracellular matrix\, notably proteoglycans\, yet the role and mechanisms of these interactions remain poorly understood. We found that Drosophila secreted semaphorins bind specifically to glycosaminoglycan (GAG) chains of proteoglycans\, showing a preference based on the degree of sulfation. Structural analysis of Sema2b unveiled multiple GAG binding sites positioned outside canonical plexin-binding site\, with the highest affinity binding site located at the C-terminal tail\, characterized by a lysine-rich helical arrangement that appears to be conserved across secreted semaphorins. In vivo studies revealed a crucial role of the Sema2b C-terminal tail in specifying the trajectory of olfactory receptor neurons. We propose that secreted semaphorins tether to the cell surface through interactions with GAG chains of proteoglycans\, facilitating their presentation to cognate receptors on passing axons.

DESCRIPTION:The dendrite’s sweet spot: Atypical glycosylation of the neuronal surface and impact on synaptic receptor trafficking\, structure and function\nLecturer: Cyril Hanus\, PhD. / The Institute of Psychiatry and Neuroscience of Paris \nThe seminar will take place on 30. 04. 2024 from 10:00 p.m. in the Turquoise Auditorium of the IEM CAS. \nAnnotation: \nLike 75% of membrane proteins\, neurotransmitter receptors and synaptic adhesion molecules are literally covered by covalently linked sugars\, N-glycans. Despite a general awareness that N-glycans may have a strong impact on protein structure and function and are altered in a number of brain pathologies\, we know surprisingly little about N-glycosylation in the brain. Our work indicates that a substantial fraction of neuronal surface proteins\, notably glutamate AMPA receptors\, are locally synthesized in dendrites and use unconventional Golgi-independent pathways to reach the neuronal surface.
