In the course of my presentation\, I will explore the cellular foundations and molecular components dictating the development of ovarian and testicular fate in avian species. Employing comparative approaches\, I will scrutinize the distinctive attributes of the ZZ and ZW chromosome sex determination system\, elucidating its role in cell-autonomous sex determination. Lastly\, I will share my recent postdoctoral findings particularly the metabolic regulation of sex differentiation.

DESCRIPTION:Lsp1 partially substitutes Pil1 function in eisosome assembly under stress conditions\nLecturer: Ing. Petra Veselá\, Ph.D. / Department of Functional Organisation of Biomembranes \nAnnotation: Eisosomes are large hemitubular structures that underlie the invaginated microdomains in the plasma membrane of various organisms and take part in the cellular stress response. In the yeast Saccharomyces cerevisiae\, they are organised by BAR-domain containing proteins Pil1 and Lsp1. While under normal laboratory conditions\, deletion of the PIL1 gene results in the inability of cells to assemble eisosomes\, deletion of LSP1 shows no apparent phenotype. DESCRIPTION:“The novel sphingolipid synthesis regulators in cell life” \nSphingolipids are bioactive lipids and critical structural components of cell membranes. Regulation of their de novo synthesis is critical for normal mammalian development and physiology. Although the ORMDL family has been identified as the gatekeeper of the serine palmitoyl transferase (SPT) activity\, our new data suggest that other components are also involved. We will present our results on new regulators of the SPT complex based on their interaction with members of the ORMDL family using genetic\, immunogenetic and mass spectrometry approaches. Our data indicate that SPT regulatory circuits are more complex than previously thought.

DESCRIPTION:„The impact of FAM84B in retinal homeostasis“ \nFam84b is an incompletely characterised protein. Its aberrant expression has been associated with various human cancers\, with a possible reported role in endosomal trafficking. We genetically ablated Fam84b expression in mice\, and extensively characterised the subsequent age-related\, degenerative phenotype in the retinas of knockout animals. The aim of the project is to explain how the lack of Fam84b unsettles retinal homeostasis and induces the presented phenotype.

DESCRIPTION:“Canonical RNAi in vivo in mice” \nCanonical RNAi\, i.e. long dsRNA-induced mRNA degradation\, is essentially not operating in vivo in mammalian somatic cells. Low efficiency of dsRNA conversion into small interfering RNA by Dicer was identified as key limiting factor for canonical RNAi in mammals. We have genetically engineered mice to express a more active Dicer variant and investigated how this impacts mouse development and RNAi activity in vivo.

DESCRIPTION:Sensorineural hearing loss in mice lacking GABAB receptor-associated KCTD12 protein\nLecturer: Mgr. Adolf Melichar / Department of Auditory Neuroscience \nAnnotation: KCTD12 protein is the auxiliary subunit of the G-protein coupled receptor for γ-aminobutyric acid (GABA-B). KCTD12 constitutively binds to the intracellular portion of GABA-B receptor and significantly modulates its structural and functional properties. KCTD12/GABA-B complexes are characterized by high expression levels in the mammalian peripheral and central auditory system\, suggesting their important role in auditory function. DESCRIPTION:„Studying the eukaryotic cilium by a single organelle approach“ \nCilia are present on the surface of many eukaryotic cells and have critical motility\, sensory\, and signalling functions. Yet\, many fundamental aspects of their biology are poorly understood. Taking advantage of the fact\, that the cilia are experimentally well accessible\, I have been developing approaches for imaging and manipulation of individual cilia. These include correlative light and electron microscopy\, blocking transport within the cilia by physical and chemical means\, as well as in vitro reconstitution approaches. In this talk\, I will discuss the development of such methods and present the advancements in our understanding of processes responsible for building and maintenance of the cilia these approaches have enabled.

DESCRIPTION:Dear Colleagues\, \nWe are pleased to announce the annual Mini-Symposium BIOCEV Regeneration III\, which will be held on 23-24th November 2023 in BIOCEV\, Vestec. \nOur symposium aims to exchange knowledge in regenerative medicine\, tissue engineering\, 3D in vitro modelling\, organoid research\, new tools and technologies in cell culture\, and many more. \nAlong with several lectures given by well-known experts in the fields\, this year\, we encourage early career researchers to contribute to our symposium and share research achievements\, exchange opinions\, or find new contacts and cooperations. DESCRIPTION:“A chance for fertility despite bad DNA breaks (AKA The effect of crossing-over rate on fertility)” \nProgrammed meiotic DNA breaks are essential for mammalian fertility but dangerous for genome integrity. DNA-binding H3-lysine-trimethyltransferase PRDM9 determines the positions of the meiotic DNA breaks\, which can be repaired as crossovers or noncrossovers. The crossover/noncrossover decision is regulated by many factors that include RING-domain E3 ligase RNF212. The dosage of the Rnf212 gene in mouse affects the crossover rate per meiotic cell and variants in the human RNF212 gene are associated with crossover rates. Deficiency for RNF212 leads to complete arrest of meiosis at the metaphase stage. The deficiency for human and rodent PRDM9 causes partial to complete pachytene arrest\, depending on unknown genetic factors. We will present results demonstrating that the dosage of Rnf212 (and thus crossover rate) affects meiotic progression and germ cell counts of semisterile Prdm9-deficient mice. Our findings offer an insight into the mechanisms and treatment possibilities of some cases of infertility. \n

DESCRIPTION:ABC transporter blood-brain barrier function in neurodegenerative and neuroinflammatory diseases – from basic research to treatment\nLecturer: Prof. Jens Pahnke\, M.D.\, Ph.D.\, E.F.N./ University of Oslo\, Faculty of Medicine / Oslo University Hospital\, Department of Pathology\, Norway \nThe seminar will take place on 20. 11. 2023 from 2:30 p.m. in the Turquoise Auditorium of the IEM CAS. \nAnnotation: \nThe current understanding of neurodegenerative diseases focusses on the deposition of toxic peptide species as larger aggregates in the brain of effected individuals. However\, patients with sporadic neurodegenerative diseases do not overexpress these peptides. Moreover\, they are hampered in removing the physiologically produced peptides\, e.g. Aβ and α-synuclein. The blood-brain barrier plays a prominent role in the active and effective removal of metabolites and in the efflux of toxic peptides from the brain into the blood stream. \nHere\, ABC transporters were discovered by us and others to have a fundamental role in the homeostasis of the brain’s environment. In 2011\, we described a new ABC transporter (ABCC1) that increases Aβ in the brain of knock-out animals by 12-14fold. Since then\, we could show that the activation of ABC transporters can be used as a diagnostic tool and a new treatment option in elderly with Alzheimer’s disease and other neurodegenerative diseases\, such as Huntington’s disease. \n1 Pahnke\, J. et al. DESCRIPTION:“Genetic architecture of Prdm9-driven reproductive isolation between mouse subspecies” \nHybrid incompatibility\, the inviability or infertility of species hybrids\, has played a central role in speciation research since Darwin’s Origin of Species. However\, the search for genes controlling hybrid sterility\, mainly using Drosophila species hybrids\, has been largely unsuccessful\, revealing a polygenic control with complex epistatic interactions. Fifty years ago\, we identified the first hybrid sterility locus\, Hst1\, in the house mouse genome\, and 14 years ago\, we positionally cloned Prdm9 as the gene responsible for Hst1-controlled hybrid sterility. \nIn this talk\, I will summarize our current understanding of the molecular genetic mechanisms that control this unique model of mammalian hybrid sterility.

DESCRIPTION:Single-cell gene expression analysis of developing craniofacial region\nLecturer: Erika Hudáčová\, M.Sc. / Department of Developmental Biology \nThe seminar will take place on 2. 11. 2023 from 2 p.m. in the Turquoise Auditorium of the IEM CAS. \nAnnotation: In vertebrates\, the skull is composed of various skeletal elements originating from cranial neural crest cells (CNCCs). CNCCs are responsible for the formation of the cranium vault\, frontonasal prominence\, upper and lower jaw\, hyoid bone\, external ear and middle ear. The morphogenesis of the craniofacial region encompasses a number of processes\, from epithelial-mesenchymal induction and mesenchymal condensation to endochondral or intramembranous ossification. However\, the molecular and genetic pathways regulating cranial cartilage shaping and growth during embryogenesis are poorly understood. \nIn our research\, we present Wnt1-Cre2\, Meis2 conditional knock-out (cKO) mice with several craniofacial bone abnormalities resembling severe human pathologies due to improper molecular mechanisms guiding mesenchymal condensation. Using single-cell RNA sequencing\, we further focus on describing the Meis2-dependent gene regulatory network involved in mesenchymal condensation. Comparison of wild-type and Meis2 cKO embryonic stages E12.5 and E13.5 reveals multiple mesenchymal cell types responsible for craniofacial formation. We have identified markers for cell type specification and positional cues in developing craniofacial regions. Furthermore\, differences identified in specific cell types governing mesenchymal condensation and endochondral ossification correlate with the phenotype present in Meis2 cKO mice. Our transcriptomics data in Meis2 cKO show elevated expression of the genes involved in ossification\, cell-cell adhesion and cell-extracellular matrix contact. Altogether\, the identification of hitherto unknown factors controlling the growth and shape of cranial bones may potentially contribute to the mitigation or prevention of human craniofacial abnormalities. \nWe look forward to your participation. \n  \n
