DESCRIPTION:ABC transporter blood-brain barrier function in neurodegenerative and neuroinflammatory diseases – from basic research to treatment\nLecturer: Prof. Jens Pahnke\, M.D.\, Ph.D.\, E.F.N./ University of Oslo\, Faculty of Medicine / Oslo University Hospital\, Department of Pathology\, Norway \nThe seminar will take place on 20. 11. 2023 from 2:30 p.m. in the Turquoise Auditorium of the IEM CAS. \nAnnotation: \nThe current understanding of neurodegenerative diseases focusses on the deposition of toxic peptide species as larger aggregates in the brain of effected individuals. However\, patients with sporadic neurodegenerative diseases do not overexpress these peptides. Moreover\, they are hampered in removing the physiologically produced peptides\, e.g. Aβ and α-synuclein. The blood-brain barrier plays a prominent role in the active and effective removal of metabolites and in the efflux of toxic peptides from the brain into the blood stream. \nHere\, ABC transporters were discovered by us and others to have a fundamental role in the homeostasis of the brain’s environment. In 2011\, we described a new ABC transporter (ABCC1) that increases Aβ in the brain of knock-out animals by 12-14fold. Since then\, we could show that the activation of ABC transporters can be used as a diagnostic tool and a new treatment option in elderly with Alzheimer’s disease and other neurodegenerative diseases\, such as Huntington’s disease. \n1 Pahnke\, J. et al. DESCRIPTION:“Genetic architecture of Prdm9-driven reproductive isolation between mouse subspecies” \nHybrid incompatibility\, the inviability or infertility of species hybrids\, has played a central role in speciation research since Darwin’s Origin of Species. However\, the search for genes controlling hybrid sterility\, mainly using Drosophila species hybrids\, has been largely unsuccessful\, revealing a polygenic control with complex epistatic interactions. Fifty years ago\, we identified the first hybrid sterility locus\, Hst1\, in the house mouse genome\, and 14 years ago\, we positionally cloned Prdm9 as the gene responsible for Hst1-controlled hybrid sterility. \nIn this talk\, I will summarize our current understanding of the molecular genetic mechanisms that control this unique model of mammalian hybrid sterility.

DESCRIPTION:Single-cell gene expression analysis of developing craniofacial region\nLecturer: Erika Hudáčová\, M.Sc. / Department of Developmental Biology \nThe seminar will take place on 2. 11. 2023 from 2 p.m. in the Turquoise Auditorium of the IEM CAS. \nAnnotation: In vertebrates\, the skull is composed of various skeletal elements originating from cranial neural crest cells (CNCCs). CNCCs are responsible for the formation of the cranium vault\, frontonasal prominence\, upper and lower jaw\, hyoid bone\, external ear and middle ear. The morphogenesis of the craniofacial region encompasses a number of processes\, from epithelial-mesenchymal induction and mesenchymal condensation to endochondral or intramembranous ossification. However\, the molecular and genetic pathways regulating cranial cartilage shaping and growth during embryogenesis are poorly understood. \nIn our research\, we present Wnt1-Cre2\, Meis2 conditional knock-out (cKO) mice with several craniofacial bone abnormalities resembling severe human pathologies due to improper molecular mechanisms guiding mesenchymal condensation. Using single-cell RNA sequencing\, we further focus on describing the Meis2-dependent gene regulatory network involved in mesenchymal condensation. Comparison of wild-type and Meis2 cKO embryonic stages E12.5 and E13.5 reveals multiple mesenchymal cell types responsible for craniofacial formation. We have identified markers for cell type specification and positional cues in developing craniofacial regions. Furthermore\, differences identified in specific cell types governing mesenchymal condensation and endochondral ossification correlate with the phenotype present in Meis2 cKO mice. Our transcriptomics data in Meis2 cKO show elevated expression of the genes involved in ossification\, cell-cell adhesion and cell-extracellular matrix contact. Altogether\, the identification of hitherto unknown factors controlling the growth and shape of cranial bones may potentially contribute to the mitigation or prevention of human craniofacial abnormalities. \nWe look forward to your participation. \n  \n
