The dendrite’s sweet spot: Atypical glycosylation of the neuronal surface and impact on synaptic receptor trafficking, structure and function

Annotation:

Like 75% of membrane proteins, neurotransmitter receptors and synaptic adhesion molecules are literally covered by covalently linked sugars, N-glycans. Despite a general awareness that N-glycans may have a strong impact on protein structure and function and are altered in a number of brain pathologies, we know surprisingly little about N-glycosylation in the brain. Our work indicates that a substantial fraction of neuronal surface proteins, notably glutamate AMPA receptors, are locally synthesized in dendrites and use unconventional Golgi-independent pathways to reach the neuronal surface. This local processing modifies protein N-glycosylation states and hence regulates receptor chemical composition and function, unravelling a previously unrecognized mechanism controlling the sensing properties and plasticity of the neuronal membrane.
